During the game, Portal, you first are just moving through the levels, but as you progress Glados tells you that you're invited to a party and will have a cake. Well, as the game advances and plotline shifts and then the cake is seen as a lie because you are seen to be killed at the end of game. When you avoid being killed and are sent to destroy Glados all over the walls in blood and other red fluids the words "HELP" and "The cake is a lie" repeat over and over.

That is what "The cake is a lie!" came from. :3
