I don't expect that you still use this method, but I have an eye where the pupil needs to be a different color from the iris. The iris is yellow, and the pupil is bright pink.

In SAI there is this brush called the Binary Tool. That is the pixeling brush in SAI.
As for Photoshop, left click the brush tool, and it'll show a small dropdown. The pencil tool is the pixeling tool in this case.

Uh, put down the base color and follow the instructions. If you really have a problem, you might want to use multiply/overlay. (If you don't know what that is, I honestly don't want to explain it, so sorry!)

Pixel is pixelly - grainy. It's not smooth. Now compare this eye to another eye made in SAI or Photoshop using the Pen tool. Difference seen.

There's a tool called the binary tool. It's made for pixels.
There's no eraser in SAI for pixeling, but there's this thing that looks like a 2x2 grid near the color boxes, right? that's for the transparent color, which basically can turn any tool into an eraser.
